摘要
目的了解体内转铁蛋白受体(TfR)mRNA表达的调节,为肝癌早期诊断和导向治疗提供依据。方法将大鼠随机分成肝硬化组和对照组,形成三种不同铁水平,分别采取部分肝切除后不同时相大鼠肝脏以及肝癌患者手术标本进行Northernblotting分析。结果无论分组,TfRmRNA表达在缺铁时最高,正常铁水平次之,高铁时最低;正常铁水平下硬化组表达高于对照组,在肝切除后不同时相TfRmRNA表达水平不同,而缺铁及高铁水平则无此变化;肝癌中癌组织表达最高,癌周组织次之,正常组织最低。结论体内调节TfRmRNA的最基本因素为铁,TfRmRNA在肝硬化及肝癌中有高表达。
Objective To understand the regulation of the expression of transferrin receptor (TfR) mRNA which provides evidence of early diagnosis of liver cancer and target-therapy. Methods Fifty rats were randomized to divide into experiment and control group and different iron level was induced by diet and repeated bleeding, rat liver specimens at 6, 12, 24h after partial hepatectomy as well as from several hepatic neoplasma patients and analys by Northern blotting. Results The expression of TfR mRNA is high at iron deficiency, low at the normal iron level and very low at iron overload regardless of the group. The expression has notable difference between cirrhosis group and control group under the normal iron level, and has time-dependent change after partial hepatectomy. The out come is not the same in iron deficiency and iron overloard. Expression is highest in liver tumor itself, second in pert-neoplasma tissue and normal liver tissue. Conclusion Iron is the most basic principle regulate factor in the expression of Tin mRNA. The exprssion of TfR mRNA is higher in the cirrhosis tissue and tumor tissue than in the normal tissue.
