目的 观察雷公藤甲素 (T10 )对平滑肌细胞 (SMCs)细胞周期进程的影响和诱导凋亡作用。方法 体外主动脉SMCs培养和流式细胞仪技术。结果 T10 与指数增殖期SMCs共育后 ,可引起核染色质浓缩、聚集 ,细胞膜内陷 ,凋亡小体形成等凋亡形态学改变。流式细胞仪分析 ,在G1峰前出现凋亡峰 ,峰值随T10 浓度增加而升高 ;G1期细胞进入S期并阻滞于S期 ;G1、M期细胞比例下降而S期升高 ;凋亡分子Fas表达明显升高。结论 T10

AIM To study the effect of triptolide (T 10 ) on the apoptosis and cell cycle of SMCs. METHODS The culture of aorta smooth muscle cells (SMCs) in vitro and flow cytometer SMCs went through three to six passages before being usingRESULTS After incubated with T 10 , exponentially proliferating SMCs changed with apoptotic features including shrinking, condensation of chromatin and formation of apoptotic body. Apoptotic peak appeared and elevated with the increasing of drug concentration. Cells of G 1 phase moved into S phase and were blocked here. The level of Fas protein increased marklyCONCLUSION T 10 can induce exponentially proliferating SMCs to apoptosis
