摘要
采用检测猪繁殖与呼吸障碍综合征病毒(PRRSV)两种不同目标蛋白抗体的间接ELISA方法对来自10个规模猪场216份血清进行检测,结果显示,包被抗原分别为核蛋白(Nucleoprotein,N)和以GP5蛋白为主的膜蛋白(membrane pro-teins,G)的两种方法检测阳性率分别为83.8%和81.9%,符合率为92.6%,Kappa值为0.74,表明两种检测方法的检测结果在给定的临界点条件下抗体检测结果具有良好的一致性。通过数据标准化,调整阴阳性判定的临界点,发现两种检测方法所得出的阳性率变化趋势相似,但检测膜蛋白的G-ELISA抗体阳性率其降幅显著高于检测核蛋白的N-ELISA。本试验初步阐明了检测猪繁殖与呼吸障碍综合征病毒抗体的两种间接ELISA方法的特点,为科学使用提供依据。
216 swine sera were collected from 10 pig farms and detected for antibody against porcine reproductive and respiratory syndrome virus（PRRSV） antibody using two commercial ELISA kits against two different viral proteins.The results showed the PRRS antibody positive rate from two ELISA kits with coating antigen of nucleoprotein（N） or membrane proteins（G） was 83.8% and 81.9%,respectively.Coincidence rate with the two ELISA kits was 92.6%,and Kappa value was 0.74.No difference was observed on PRRS antibody rates by two ELISA kits using provided positive/negative threshold.Through adjusting the threshold value by standardizing the data,the trend of positive rates in two assays was similar,and the rate showed the decline with the increase of threshold value.However,the modification of positive rate of G-ELISA assay is greater than that of N-ELISA assay,by changing the threshold.This study showed that the characteristics of the two ELISA assays for detecting PRRSV antibody are not similar when using for different purposes.
