化浊解毒方对裸鼠人胃癌原位移植瘤VEGF与COX-2的影响 被引量:3

Effects of Huazhuo Jiedu Recipe on VEGF and COX-2 Expression in Human Gastric Cancer Transplanted Tumor in Nude Mice

摘要 目的:研究化浊解毒方对裸鼠人胃癌原位移植瘤组织中血管内皮生长因子(VEGF)、环氧合酶(COX)-2表达的影响。方法:BALB/c裸鼠40只,人胃癌原位移植瘤SGC-7901细胞株移植于裸鼠皮下以复制裸鼠人胃癌原位移植瘤模型,复制模型7d后,40只模型裸鼠随机分为4组,即模型(等容生理盐水,灌胃,每天1次)、中药(化浊解毒方0.4mL,灌胃,每天1次)、化疗(卡铂5mg·kg-1,腹腔注射,每3天1次)、联合用药(化浊解毒方0.4mL,灌胃,每天1次+卡铂5mg·kg-1,腹腔注射,每3天1次)组,连续4周。裸鼠每周称重2次,测量肿瘤2次。末次给药后2周末观察瘤体积与瘤重的变化,通过免疫组化法检测各组VEGF、COX-2的表达。结果:与模型组比较,中药、联合用药、化疗组抑瘤率显著增加,VEGF、COX-2的表达显著降低(P<0.05);而联合用药组优于化疗组。结论:化浊解毒方可降低模型裸鼠VEGF、COX-2的表达,联合用药疗效更佳。 OBJECTIVE:To study the effects of Huazhuo jiedu recipe on the expressions of vascular endothelial growth factor(VEGF) and cyclooxygenase-2(COX-2) in human gastric carcinoma tissues in nude mice underwent orthotopic transplantation.METHODS:40 BALB/c nude mice were collected.We established human gastric cancer orthotopic transplantation/metastatic model of nude mice by transplanting human gastric cancer cell lines(SGC-7901) subcutaneously in nude mice.After modeling for seventh days,40 tumor-bearing nude mice were randomly divided into 4 groups,i.e.model group(constant volume normal saline,i.g.once a day),Traditional Chinese medicine group(Huazhuo jiedu recipe 0.4 mL,i.g.once a day),chemotherapy group(carboplatin 5 mg·kg-1,i.p.every 3 days),combined medication group(Huazhuo jiedu recipe 0.4 mL,i.g.once a day+carboplatin 5 mg·kg-1,i.p.every 3 days) for consecutive 4 weeks.Nude mice were weighed 2 times every week,and tumor was measured 2 times.The changes and metastasis of tumor volume and tumor weight were observed 2 weeks after the last administration,and immunohistochemical method was used to detect the expressions of VEGF and COX-2.RESULTS:The tumor-suppressing rates were higher,the expressions of VEGF and the tumor-suppressing rates were counted,and COX-2 in traditional Chinese medicine group,combined medication group and chemotherapy group were lower than that in model group significantly(P0.05);that of combined medication group was better than that of chemotherapy groups.CONCLUSION:Huazhuo jiedu recipe can reduce the expression of VEGF and COX-2 in model rats,and therapeutic efficacy of drug combination is better than others.
