The role of interleukin-22 in hepatitis and hepatocellular carcinoma

摘要 IL-022是2000年才发现的细胞因子,主要由Th22细胞分泌,以皮肤,胰腺,肝脏等组织细胞为主要靶细胞。IL-22可激活JAK—STAT3信号通路,诱导组织细胞的多种基因表达上调,这些基因涉及组织重塑、血管生成、趋化细胞因子生成、纤维化形成等。因此,IL-22在感染免疫、组织修复、肝炎产生和肝癌形成中起重要免疫病理作用。大量的文献报道IL-22在刀豆蛋白、四氯化碳等介导的T细胞肝炎中起着减轻细胞损伤、促进细胞增殖和加速组织修复等保护性作用,但也有些证据表明IL-22在HBV感染中促进炎症发生和诱导肝癌形成。故进一步阐明IL-22在肝炎和肝癌中的作用对肝炎的治疗和肝癌的预防有重要意义。 Interleukin-22 (IL-22), a novel cytokine identified in 2000, was secreted by T helper 22 ( Th22 ) cells. It is now known that the target cells of IL-22 mainly include epithelial cells, pancreatic cells and hepatocytes. IL-22 exerts its functions by activation of JAK-STAT3 signal pathway, followed by up-regulation a variety of genes which associate with tissue repair, angiogenesis, ehemokine generation and fibrosis. So IL-22 plays an important role in infection immunity, tissue repair, liver inflammation and tumorigenesis. Regarding to the liver, most of the evidence supported that IL-22 plays a protective role in preventing hepatocyte damage, and also some studies show that IL-22 acts as a pro-inflammatory and tumor-promoting factor in some disease processes. The purpose of this review was to describes the function of Interleukin-22 in hepatitis and hepatoeel- lular carcinoma.
