摘要
为阐明狂犬病病毒(RV)不同毒力株感染鼠脑后基因表达的差异,进一步揭示RV感染和机体抗感染应答的分子机制。本试验应用差异显示技术分析了正常鼠脑悬液、狂犬病病毒SRV9弱毒株及BD06街毒株感染鼠脑48h的基因水平变化。结果显示,与对照组相比,SRV9与BD06组有4对共同上调表达差异片段;与其它两组比较,SRV9组有2个基因上调表达,BD06组存在1个上调表达基因。这些差异基因体现了宿主细胞对RV感染的应答模式以及不同毒株感染间的差异,为深入研究RV致病机理奠定了基础。
To clarify the differences of gene impre ssion in mouse brain after infected by virulent and attenuated rabies virus stra ins,further to reveal the mechanism of virus infection and hosts' anti-infecti on response,this study co mpared the gene expression in mouse brains infected by attenuated(SRV9) and vir ulent(BD06) RV strain after 48 hours by using the techn ic DDRT-PCR.The result showed that the expression of four genes was up-regulat ed in both BD06 and SRV9 groups compared with control group;two strips showed t he higher gene expression in SRV9 group than in BD06 group and control grtoup;a nd one strip was found up-regulated expression in BD06 group compared with the oth er groups.These differential gene expression showed that there were differences among the reaction models when host cells were infected by RV,and the suitability of different virulence RV strains infection,which could be of significant sense in investgating the pathogenesis of rabies virus.
