摘要
目的研究1,25-二羟维生素D3[1,25(OH)2D3]对胃癌SGC-7901细胞增殖和细胞周期的影响,以及对细胞因子TNF-αmRNA及蛋白表达的影响。方法用不同浓度1,25(OH)2D3处理胃癌SGC-7901细胞,采用MTT法检测细胞增殖情况;流式细胞仪检测细胞生长周期;RT-PCR和Western blotting分别检测TNF-α在mRNA和蛋白水平的表达情况。结果 1,25(OH)2D3对胃癌SGC-7901细胞增殖有抑制作用(P<0.05),并呈时间剂量依赖性。经1,25(OH)2D3处理72 h后细胞周期出现向G0/G1期移行的动力学改变(F=9.81,P<0.05)。TNF-αmRNA及蛋白的表达水平均呈剂量依赖性减少(P<0.05)。结论 1,25(OH)2D3可抑制胃癌细胞增殖,且与其抑制TNF-α的表达可能有相关性。
Objective To investigate the effect of 1,25(OH)2D3 on cell proliferation and the expression of tumor necrosis factor-α(TNF-α) in human gastric carcinoma SGC-7901 cells.Methods SGC-7901 cells were treated with 1×10-6,1×10-7,1×10-8,and 1×10-9 mol/L 1,25(OH)2D3 for 24,48,72 and 96 h.The cell proliferation was measured by MTT assay,and the cell cycle changes were analyzed using flow cytometry.RT-PCR and Western blotting were used to determine the expression of TNF-α mRNA and protein,respectively.Results 1,25(OH)2D3 significantly inhibited SGC-7901 cell proliferation(P0.05) in a time-and dose-dependent fashion.Treatment with 1,25(OH)2D3 for 72 h caused significant cell cycle arrest at G0/G1 phase(F=9.81,P0.05) and dose-dependently inhibited the expression of TNF-α at both mRNA and protein levels in SGC-7901 cells(P0.05).Conclusion The inhibitory effect of 1,25(OH)2D3 on SGC-7901 cell proliferation is probably associated with the down-regulation of TNF-α expression.
