摘要
目的观察慢性阻塞性肺疾病(COPD)患者肺组织的丙二醛(MDA)、白细胞介素8(IL-8)与肿瘤坏死因子α(TNF-α)水平,探讨其在COPD发病中的作用。方法采用酶联免疫吸附试验(ELISA)检测COPD患者中的吸烟者、曾经吸烟者、非吸烟者及非COPD患者中的吸烟者、曾经吸烟者、非吸烟者的肺组织MDA、IL-8与TNF-α含量,采用肺组织匀浆的肌酐浓度进行校正。结果COPD患者肺组织MDA、IL-8和TNF-α水平明显高于非COPD患者(P<0.01)。无论罹患COPD与否,吸烟者肺组织MDA、IL-8和TNF-α水平均明显高于非吸烟者(P<0.05);COPD患者中的吸烟者肺部MDA、TNF-α水平显著高于曾经吸烟者(P<0.05),但二者肺部IL-8水平无明显差异(P>0.05);非COPD患者中的吸烟者肺组织TNF-α水平显著高于曾经吸烟者(P<0.05),但肺组织MDA、IL-8水平差异无统计学意义(P>0.05)。结论长期吸烟可引起肺组织MDA、IL-8和TNF-α水平异常升高,可能在COPD的发病中起重要作用。
Objective To observe the levels of malonaldehyde(MDA),i nter leukin-8(IL-8) and tumor necrosis factor-α(TNF-α) in lung tissues of sub ject s with or without chronic obstructive pulmonary disease(COPD),and investigate their roles in the the pathogenesis of COPD.Methods The cont ent of MDA,IL-8 and TNF-α in lung tissues of smokers with COPD(n=9),ex-smokers with COP D(n=8),non-smokers with COPD(n=7),healthy smokers(n=9),healthy ex-smok ers(n=8) and healthy non-smokers(n=6) was measured with enzyme-linked immunosorben t ass ay(ELISA) and corrected by creatinine.Results The levels of MDA,IL-8 and TNFα in lung tissues of the COPD patients were significantly higher than those in the heal thy subjects(P0.05),which were also significantly higher in the smokers when comp ared with the non-smokers(P0.05),whether suffering from COPD or not.In the COPD pa tients, not the levels of IL-8 but MDA and TNF-α in lung tissues of the smokers were sig ni ficantly higher than those in the ex-smokers(P0.05);whereas in the healthy case s,no statistical significance was revealed between the smokers and the ex-smokers on the leve l s of MDA and IL-8 in lung tissues except TNF-α(P0.05).Conclu sion The abnorm al increase of MDA,IL-8 and TNF-α in lung tissues caused by chronic smoking may play an important role in the the pathogenesis of COPD.
