摘要
目的观察灯盏花素对缺氧/复氧大鼠心肌细胞凋亡相关基因Janus激酶(JAK2)mRNA及其蛋白表达的影响,试图在细胞水平上为临床应用灯盏花素减轻心肌缺血再灌注损伤提供理论依据。方法取原代培养的乳鼠心肌细胞,随机分为4组:正常对照组,模型组,灯盏花素2个剂量组(25 mg/L,50 mg/L),其中模型组和灯盏花素2个剂量组进行缺氧2 h,再复氧,并于复氧4 h后,分别采用RT-PCR和Western blot检测大鼠心肌细胞JAK2基因mRNA及其蛋白的表达。结果模型组大鼠心肌细胞JAK2基因mRNA和蛋白条带平均光密度值均较正常对照组增加(P<0.01);与模型组相比,灯盏花素2个剂量组大鼠心肌细胞JAK2基因的mRNA和蛋白条带平均光密度值均降低(P<0.05,0.01)。结论灯盏花素可抑制缺氧/复氧大鼠心肌细胞凋亡相关基因JAK2 mRNA,进而抑制了JAK2蛋白表达,从而抑制缺氧/复氧引起的大鼠心肌细胞凋亡的作用。
AIM To investigate the effects of Breviscapine Injection on the expression of JAK2(Janus kinase 2) mRNA and its protein in cardiomyocytes of rats with oxygen defect and resupply,and to provide evidence for the utilization of breviscapine against the ischemia reperfusion injury.METHODS The cultured neonatal rat cardiomyocytes were divided into four groups: normal control group,model group,two dose breviscapine groups(25 mg/L,50 mg/L).Except the normal control group,cells collected at oxygen defect for 2 h,oxygen resupply for 4 h were determined with RT-PCR and Western blot,to measure the expression of mRNA and protein of JAK2 gene.RESULTS Compared with the normal control group,the mean optic density(MOD) of mRNA and protein expression of JAK2 gene in cardiomyocytes of rats increased obviously in the model group(P〈0.01).MOD of mRNA and protein expression of JAK2 gene in cardiomyocytes of rats decreased obviously in the Breviscapine Injection group(P〈0.05,0.01);CONCLUSION Breviscapine Injection can inhibit the mRNA expression of JAK2 gene and the protein expression of JAK2 gene further affects the apoptosis of cardiomyocytes of rats.
