Determination of 5-Aminolevulinic Acid by Pre-column Derivatization HPLC-FD

摘要 建立了柱前衍生化HPLC-荧光法测定5-氨基酮戊酸。以荧光胺为荧光衍生化试剂,使用C18柱,以乙腈-0.1%三氟乙酸溶液(30︰70)为流动相,激发波长和发射波长为398和480 nm。5-氨基酮戊酸在0.047 5~47.5 g/ml浓度范围内线性关系良好。回收率为99.2%,RSD为1.46%。 A pre-column derivatization HPLC-FD method was established for the determination of 5-aminolevulinic acid.Fluorescamine was used as the derivatizing agent.A C18 column was used with the mobile phase of acetonitrile-0.1% trifluoroacetic acid solution(30︰70).The excitation and emission wavelengths were 398 and 480 nm.The calibration curve of 5-aminolevulinic acid was linear in the concentration range of 0.047 5-47.5 μg/ml.The average recovery was 99.2%,with RSD of 1.46%.
