富含12-MTA的嗜碱芽孢杆菌X1的鉴定及其脂肪酸组成分析 被引量:2

摘要 12-甲基十四烷酸(12-MTA,anteiso-C15∶0)具有显著的抗肿瘤活性。以重度盐碱土中分离的产芽孢细菌X1为研究对象,通过16S rDNA系统发育分析和表型特征确定其分类地位,通过细胞裂解、脂肪皂化、盐酸催化的甲酯化和萃取等步骤制备X1菌体总脂肪酸甲酯衍生物,GC-MS分析其组成,结合称重法测定X1菌体总脂肪酸甲酯衍生物净重。结果表明:菌株X1为芽孢杆菌属(Bacillus)的成员,其16S rDNA相似性数据显示其为芽孢杆菌属内的一个新种。GC-MS分析确定X1菌体主要含有iso-C14∶0、C14∶0i、so-C15∶0和anteiso-C15∶0 4种脂肪酸,其中anteiso-C15∶0(12-MTA)比例占到X1菌体总脂肪酸含量的76.2%,含量占X1菌体干重的4.47%。 12 - methyltetradecanoic acid ( 12 - MTA, anteiso - C15:0 ) possesses notable antitumor activity. The spore - producing bacterium X1 isolated from severe saline - alkali soil was taken as research ob- ject, and its taxonomic position was determined by 16S rDNA -based phylogenetic analysis and phenotypic characteristics. The total fatty acid methyl ester derivatives of X1 were prepared through cell lysis, fat saponification, HC1 -catalyzed methyl esterification and extraction, and their compositions were analyzed by GC - MS. The net weight of total fatty acid methyl ester derivatives of X1 was determined by weighing method. The results showed that the strain X1 was a member of genus Bacillus, and the 16S rDNA similarity data showed it should be a novel species within the genus Bacillus. GC - MS analysis determined that fatty acids of X1 mainly contained four kinds of fatty acids which were iso -C1,0 ,C14:o ,iso- C15:0 and anteiso - C15:0, of which anteiso - C15:0( 12 - MTA) was 76.2% of the total fatty acid content of X1 and 4.47% of X1 dry weight.
