CpG寡核苷酸诱导人外周血单核细胞抗膀胱肿瘤BIU-87细胞的杀伤作用

摘要 目的研究A型与B型CpGODN对人外周血单个核细胞(PBMC)的增殖,细胞因子的释放和诱导人PBMC抗膀胱肿瘤细胞作用。方法用A型、B型CpGODN和NoCPGODN刺激正常人外周血PBMC,MTT测定细胞的增殖,定量ELISA检测人干扰素-α含量,LDH法检测CpGODN诱导后人PBMC对膀胱肿瘤细胞的杀伤作用。结果A型与B型CpGODN对人PBMC增殖的刺激作用明显高于对照组;CpGODN2216组IFN—α的浓度高于CPGODN2006组;CpG寡核苷酸作用后,人PBMC对BIU-87膀胱肿瘤细胞的杀伤活性明显升高。结论A型与B型CpGODN对人PBMC具有免疫诱导作用,可刺激人PBMC增殖并释放IFN—α,诱导免疫细胞对膀胱肿瘤细胞杀伤,增强宿主抗膀胱肿瘤细胞作用。 Objectives The study was designed to investigate the function of the effect of Type A and B CpG oligodeoxynucleotides on the differentiation, cytokine secreted by PBMC, cytotype change of B cells. Methods To test lymphocyte proliferation by MTT method after human PBMC was incubated with type A and B CpG ODN; To measure cytokine IFN - α release by ELISA on human PBMC induced by CpG ODN;Analysis B cell surface marker of human PBMC by flow cytometry. Results Both type A and B CpG ODN could induce proliferation of PBMC. Induct effection of type A is significantly higher than type B. Type A CpG ODN strongly induce PDCs to secrete IFN -α. Type B CpG ODNs induce less IFN - α and primarily activate B ceils, causing them to up - regulate CI)69. Conclusions CpG ODN could have immune induction of human PBMC and up - regulate immune function.
