中药止痛巴布贴对骨癌痛大鼠痛行为及脊髓背角c-fos表达的影响 被引量:2

Effect of Pain-Relieving Plaster on Pain Behaviors and c-Fos Expression in Spinal Dorsal Horn in Rats with Bone Cancer

摘要 目的:观察中药止痛巴布贴对骨癌痛大鼠痛行为及脊髓背角神经节c-fos表达的影响。方法:108只180~220g雌性Wistar大鼠随机分为3组,分别为空白对照组(Con组,n=24)、假手术组(Sham组,n=24)、骨癌痛模型组(Ca组,n=60),3组分别于手术前2天、术后7、14、21日测定机械痛阈(MWT)和热痛阈(TWL)。于术后7天确认造模成功后,将成功的模型鼠随机分为两组,即模型组(Ca组,n=25)和中药止痛巴布贴组(CM组,n=18)。术后7、14和21天各组处死大鼠(n≥8),取大鼠脊髓腰椎L4-6膨大处,用免疫组化方法测定脊髓背角c-fos的变化。结果:MWT、TWL:术后Sham组、Ca组、CM组均低于基础值;术后7天,Sham组与基础值、Con组差异无统计学意义(P>0.05),Ca组、CM组则低于基础值、Con组和Sham组(P<0.05);术后14、21天Ca组与基础值、Con组和Sham组统计学差异逐渐增大(P<0.01),而CM组则差异逐渐缩小(P>0.05)。脊髓背角神经Fos表达:Ca组阳性神经元数目增加,术后7、14、21天与Con组和Sham组比较有统计学差异(P<0.05);CM组于术后14、21天与Con组和Sham组比较统计学差异逐渐缩小(P>0.05)。结论:中药止痛巴布贴对骨癌痛有比较明显的镇痛作用,其作用是通过影响脊髓背角神经细胞c-fos基因的表达而产生的。 Objective:The impacts of Chinese Traditional Medicine Pain-Relieving Plaster on the pain behaviors and the influence of the expression of c-fos in the spinal cord dorsal root ganglion of rats with bone cancer were observed.Method:108 female Wistar rats(180~220g)were randomly divided into 3 groups,blank control group(group Con,n=24),sham operation group(group Sham,n=24),bone cancer pain model group(group Ca,n=60).2 days before and 7,14,21 days after operation,mechanical pain threshold(MWT)and thermal pain threshold(TWL)in the 3 groups were tested.On the 7th day postoperatively confirmed after injecting,the success of the model rats were randomly divided into two groups,namely the model group(group Ca,n=28)and Chinese Traditional Medicine Pain-Relieving Plaster group(group CM,n=25).After 7 days,14 days and 21 days rats in each group were sacrificed(n≥8),rat spinal cord lumbar L4-6 was modified by immunohistochemical method,the changes of Fos in spinal dorsal horn were observed.Result:(1)MWT、TWL:the levels in Postoperative Sham group,Ca group,CM group were lower than the base value;7 days after operation,the levels in Sham group and Con group,combined with base value,the difference was not statistically significant(P0.05);the levels in Ca group,CM group were lower than the base value and that in Con group and Sham group(P0.05).But levels in group CM after 14,21 days,combined with the base and that in Con group and Sham group,the difference was increased(P0.05);after 14,21 days,the difference between group Ca and basic value,Con group,Sham group were statistically significant increased gradually(P0.01),while in CM group the differences become smaller(p0.05).(2)Spinal cord dorsal horn neuronal Fos expression:positive neurons in group Ca increased,after 7,14,21 days,compared with in group Sham there was statistical difference(P0.05);after 14,21 days,comparison between group CM group and Con and group Sham were statistically different gradually(P0.05).Conclusion:Chinese Traditional Medicine Pain-Relieving Plaster has obvious analgesic effect,its role is through the effects on spinal dorsal root ganglion of the expression of Fos protein.
