芹菜素对同型半胱氨酸诱导的人脐静脉内皮细胞氧化损伤的保护作用 被引量:2

摘要 目的:探讨芹菜素(Apigenin)对同型半胱氨酸氧化损伤的人脐静脉内皮细胞的保护作用。方法:体外培养内皮细胞,将细胞分为7组,即空白对照组(control)、溶剂对照组(DMSO)、Hcy氧化损伤组(Hcy)、氧化损伤加入槲皮素对照组(quercetin+Hcy)、氧化损伤加入芹菜素低、中、高浓度组(Apigenin-L+Hcy、Apigenin-M+Hcy、Apigenin-H+Hcy)。将5mmol/L Hcy作用于加入槲皮素及不同浓度芹菜素预培养6h的内皮细胞,继续培养24h,以细胞毒四唑盐(MTT)比色试验,乳酸脱氢酶(LDH)、谷胱甘肽过氧化酶(GSH-px)、丙二醛(MDA)、活性氧,NADPH酶活性、流式细胞凋亡率为检测指标。结果:芹菜素呈剂量依赖性降低同型半胱氨酸对内皮细胞活力的影响,降低MDA、LDH量,增加培养液中GSH-px活性,并显著减少活性氧、NADPH酶活性和细胞凋亡数量,各指标差异有显著性(P<0.01)。结论:芹菜素可保护和修复同型半胱氨酸诱导的内皮细胞的损伤,其作用可能与抗氧化、抑制内皮细胞凋亡有关。 Objective:To study the effect of apigenin on human umbilical vein cells(HUVECs)injuries induced by homocysteine.Methods:HUVECs were cultured in vitro and divided into seven groups:control group;solvent group,homocysteine group,quercetin plus homocysteine,apigenin-L、M、H plus homocysteine group respectively.Endothelial cells were incubated with 5 mmol/L homocysteine for 24 hours in the absence or presence of various concentrations of apigenin and quercetin.Then the apoptotic indexes were detected by flow cytometric.Meanwhile,the amount of Malondialdehyde(MDA),the levels of ROS,the activities of GSH-px and NADPH oxidase were measured.Results:Incubation of HUVECs with Hcy for 24h elicited the increaseing rate of apoptotic cells,the release of LDH,ROS levels and the activty of NADPH.Pretreatment of Apigenin with 1-50 μmol/L decreased these apigenin-induced changes in a concentration-dependent manner(P0.01).Conclusion:Apigenin has a protective effect against endothelial cells injuries induced by homocysteine,which is probably via anti-oxidation and inhibiting the apoptosis of endothelial cells.
