五子衍宗方对生精障碍模型大鼠睾丸TGF-β_1/Smads信号转导通路的影响 被引量:9

Effect of Wuzi Yanzong on Expression of TGF-β1 Signaling Pathwayin Testis of Aspermatogenesis Rats

摘要 目的:观察五子衍宗方对生精障碍模型大鼠睾丸TGF-β1/Smads信号转导通路的影响,探讨其治疗男性不育症的作用机制。方法:采用随机数字表法将50只雄性SD大鼠分为正常对照组、模型组、甲基睾酮组和五子衍宗方低、高剂量组,每组10只。除正常对照组外,其余各组大鼠按醋酸棉酚50mg/kg体质量灌胃给药,制备生精障碍模型。正常组和模型组大鼠灌服蒸馏水,甲基睾酮组大鼠灌服甲基睾酮混悬液,五子衍宗方低、高剂量组大鼠分别给予低、高浓度(含1.75g、3.5g生药/mL)五子衍宗方混悬液灌胃,每天1次,连续30天。光镜下观察大鼠睾丸组织结构变化,免疫组化法检测大鼠睾丸组织TGF-β1、Smad2、Smad4蛋白表达量。结果:棉酚可造成大鼠睾九曲细精管萎缩、退化,管壁变薄,生精细胞减少,排列紊乱,精子发生障碍;睾丸组织TGF-β1表达上调(P<0.01),Smad2、Smad4表达下调(P<0.01)。五子衍宗方能显著下调大鼠睾丸TGF-β1表达(P<0.01),上调Smad2、Smad4表达(P<0.01),拮抗棉酚造成的大鼠睾丸组织结构损伤。结论:通过调控TGF-β1/Smads信号转导通路,改善大鼠睾丸的生精功能,可能五子衍宗方治疗不育症的作用机制之一。 objective: To investigate the effect of Wuzi Yanzong on the TGF-β1/smads signaling pathway in testis of model of aspermatogenesis and to study the mechanism about it treating male's infertility.Method: Altogether 50 SD rats were randomly divided into five groups: normal group,model group,Methyltestosterone(MT),low and high dosage Wuzi Yanzong groups.Apart from the control group,these rats were treated with 50mg/kg gossypol acetic acid(GAA) for 30 days to establish the model of aspermatogenesis.The control and model group were gived distilled water,MT group gived MT suspl.,low and high dosage group gived Wuzi Yangzong suspl.(1.75g,3.5g/mL) separately.and observations made on the structure of testis using the optical microscope,expression of TGF-β1、Smad2、Smad4 protein detected.Result: Gossypol could induce convoluted tubules trophied,degeneration,spermatogenic cells decreased,the arrangement of spermatogenic cells in disorder,the sperm count significantly decreased;The expression of TGF-β1 protein increased significantly(P0.01),Smad2 and Smad4 decreased significantly(P0.01).Wuzi Yanzong could decrease The expression of TGF-β1 protein(P0.01),increase The expression of Smad2 and Smad4 protein significantly(P0.01),relieve injury of the structure of testis by gosspol.Conclusion: Wuzi Yanzong can regulate the TGF-β1 signaling pathway,improve the maturation of testis,that is one of the mechanism for Wuzi Yanzong treating male's infertility.
