益气活血中药和辛伐他汀西药对球囊损伤后动脉粥样硬化家兔血管内皮功能的影响 被引量:8

摘要 目的:通过建立家兔颈总动脉球囊损伤动脉粥样硬化模型,观察益气活血中药和辛伐他汀西药对家兔血管内皮功能的影响,研究益气活血复方抗动脉粥样硬化的作用机制。方法:实验选取普通雄性新西兰大白兔38只,随机分成正常对照组(8只),辛伐他汀组;益气活血复方组;模型组(均10只)。适应性喂养1周后,后3组行颈总动脉球囊损伤术。术后正常组给予普通饲料,其余各组给予高脂饲料。各组分别于第0周、4周、8周空腹耳中央动脉采血测定血浆NO含量,内皮素-1水平,并取颈总动脉组织观察形态学变化。结果:球囊损伤后第4周末,模型组兔血浆NO含量较正常对照组明显降低(P<0.01),辛伐他汀组和益气活血复方组兔血浆NO含量较同期模型组明显升高(P<0.01),其中益气活血复方组兔血浆NO水平升高较辛伐他汀组更为明显(P<0.05)。球囊损伤后第4周末,模型组家兔血浆ET-1水平较正常对照组明显上升;与模型组比较,辛伐他汀组及益气活血复方组血浆ET-1水平均显著下降(P<0.05),但两组间无显著性差异(P>0.05)。辛伐他汀组及益气活血复方组颈总动脉组织形态学改变较同期模型组有明显改善。结论:益气活血复方可促进NO合成和分泌,抑制ET-1表达,有利于恢复NO/ET-1平衡,能明显改善动脉粥样硬化组织形态的变化,提示益气活血复方可能具有的内皮恢复及改善功能在防止AS发生中有积极作用。 Objective:To observe the influence of Qi-supplementing-and-blood-activating compound formula on endothelial function in rabbit with arthrosclerosis and to research the mechanism.Methods:38 New Zealand male rabbits were divided into 4 groups at random : normal group(n=8),model group(n=10),Qi-supplementing-and-blood-activating group(n=10),and Simvastatin medicine group(n=10).The models were established by injuring carotid arteries.After injuring carotid arteries.,normal group was fed with normal forage and the others were fed with high fat forage.The NO and ET-1 levels were detected before and after 4、8 weeks therapy.At the 8th weekend,rabbits were sacrificed and the left and right common carotid arteries were taken and observed by transmission electron microscope.Results:After injuring carotid arteries 4 weeks,the NO level in model group was decreased gradually(P0.01).Compared with the model group,the NO levels of Qi-supplementing-and-blood-activating group and the Simvastatin medicine group were increased obviously with significant difference(P0.01),and it increased greatly in Qi-supplementing-and-blood-activating group than that in the Simvastatin medicine group(P0.05).The ET-1 level in model group was increased gradually(P0.01).Compared with the model group,the ET-1 levels in Qi-supplementing-and-blood-activating group and the Simvastatin medicine group were descended obviously with significant difference(P0.05),and there was no difference between two treatment groups(P0.05).It was confirmed in electron microscope that the Qi-supplementing-and-blood-activating compound formula and Simvastatin medicine could improve the ultrastructue of arteria carotis interna.Conclusion:The Qi-supplementing-and-blood-activating compound formula could increase the NO level and decrease ET-1 level,benfiting for keeping balance of NO/ET-1,and improve the change of arthrosclerosis obviously.It also could reduce arthrosclerosis.
