摘要 采用Ca2 + 、Mn2 + 和地中海拟无枝菌酸菌U 32共培养的方法 ,通过滤纸法检测抗生素含量和离体磷酸化反应检测激酶活性 ,研究Ca2 + 和Mn2 + 对地中海拟无枝菌酸菌U 32次生代谢的影响。低浓度Ca2 + 和Mn2 + 均能促进地中海拟无枝菌酸菌U 32的生长 ;随着离子浓度增高 ,促生长效果下降 ;浓度太高 ,则表现为抑制作用。 5mmol/LCa2 + 对地中海拟无枝菌酸菌U 32的色素绝对产量增加效果最好 ,而对其相对于菌体的色素量则以 5mmol/LMn2 + 的效果最好。 0 5mmol/LCa2 + 对地中海拟无枝菌酸菌U 32的抗生素合成有很好的作用 ,而Mn2 + 有一定的抑制作用。对地中海拟无枝菌酸菌U 32的无细胞提取液进行离体磷酸化反应检测 ,发现Ca2 + 能促进其激酶的活性 ,而Mn2 + 则相对抑制了某些激酶活性。 The effect of Ca 2+ and Mn 2+ on secondary metabolism of Amycolatopsis mediterranei U 32 were studied using co culture method of filter paper slice and phosphorylation reaction in vitro to detecting antibiotics content and protein kinase activities.The cell growth and pigment production were enhanced through adding optimum concentration of Ca 2+ and Mn 2+ .The cell growth and antibiotics production were inhibited by 1 mmol/L Mn 2+ ,but the pigment and antibiotics production were improved by 0 5 mmol/L Ca 2+ ,which could be implemented in large scale fermentation production of rifamycin in Amycolatopsis Amditerranei U 32.The phosphorylation reaction in vitro using labeled r [p 32 ] ATP with cell free extraction from mycelia indicated that the kinase activities could be affected by Ca 2+ and Mn 2+ .The result showed that the kinase activities were inhibited by Mn 2+ and enhanced by Ca 2+ .
