摘要 目的通过测定水分、总灰分、酸不溶性灰分、生物碱薄层鉴别及含量测定来研究苦豆子药材的质量控制。方法用碘化铋钾试液显色的薄层反应法鉴别生物碱,按《中国药典》规定方法检测苦豆子的水分、总灰分、酸不溶性灰分,采用高效液相色谱(HPLC)法测定氧化苦参碱、氧化槐果碱的含量。结果建立了生物碱薄层鉴别方法及氧化苦参碱、氧化槐果碱高效液相色谱(HPLC)测定方法;确定了水分、总灰分、酸不溶性灰分的含量限度,根据测定结果苦豆子药材水分在5%~7%之间为适宜,总灰分在2.0%~3.5%,酸不溶性灰分在0.2%~0.3%之间为适宜。结论测定苦豆子药材的水分、总灰分、酸不溶性灰分、生物碱的薄层鉴别及含量测定,为了建立苦豆子药材质量控制提供了奠定基础。 Objective To study medicine quality control through the determination of moisture content, to tal ash, acid-insoluble ash, TLC identification and content determination. Methods Alkaloids was identi fled by thin layer chromatography protisite reaction with bismuth potassium iodide reagent. The contents of moisture, total ash acid-insoluble ash were detected according to standards stated in Chinese Pharmacopoeia. The contents of oxymatrine, oxysophocarpine weredetermined by high pressure/per formance liquid chromatography (HPLC). Results TLC identification of alkaloids and HPLC determina tion of oxymatrine, oxysophocarpine were established preliminarily. Limitation of moisture content, ash content were defined according to the determination result of Sophora alopecuroids water in 5%~7%, to-tal ash in 2%~ 3. 5%, acid insoluble ash in 0. 2%~3% was suitable. Conclusion Determination of Sophora alopecuroides water, total ash, acid-insoluble ash, TLC identification and content determinationof alkaloids in Sophora alopecuroides can establish a quality control of medicinal provides foundation.
