不同针刺强度对急性脑缺血再灌注大鼠TNF-α、IL-1β含量的影响 被引量:8

The influence of TNF-α and IL-1β by different electroacupuncture intensity on acute cerebral ischemia reperfusion in rats

摘要 目的探讨不同针刺强度对急性脑缺血再灌注大鼠肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量的影响。方法将44只SD雄性大鼠随机分成正常组(6只)、假手术组(8只)、模型组(30只),模型组又随机分为模型对照组、弱电针组、强电针组,每组8只,另6只造模备用。采用改良线栓法建立大鼠局灶性脑缺血(MCAO)再灌注模型,针刺"百会"、"大椎"穴,接电针仪:疏密波,频率2~15Hz,调整不同电流参数,强电针参数:以肢体震颤或肌肉出现轻微强直收缩为度,电流强度4~7mA;弱电针参数:以胡须微动或肌肉轻度跳动为度,电流强度1~3mA。24h后取材,用酶联免疫吸附试验(ELISA)检测各组血清中TNF-α、IL-1β含量并进行统计学分析。结果与正常组、假手术组比较,模型对照组TNF-α,IL-1β含量差异有统计学意义(P<0.01);与模型对照组比较,各电针组能明显降低模型大鼠血清TNF-α,IL-1β含量(P<0.01~0.05);与弱电针组比较,强电针组对TNF-α、IL-1β的调节作用显著(P<0.05)。结论不同针刺强度在调节急性脑缺血再灌注大鼠炎性细胞因子TNF-α、IL-1β含量有差异,强刺激优于弱刺激。 Objective To explore the effects of different intensities of electroaeupuncture to the contents of TNFα and IL-1β on acute cerebral ischemia reperfusion in rats. Methods A total of 44 male SD rats were randomly divided into normal group (n=6), sham operation group (n=8), model group (n=30), model group was randomly divided into model compare group (n=8), weaker electroacupuncture group (n=8),stronger electroacupuncture group (n= 8). Established the focal cerebral ischemia reperfusion model (MCAO) by modified suture method in rats, and given Electroacupuncture "Baihui","dazhui": sparse- density wave, frequency of 2~15 Hz, adjustment of different current parameters, strong Electroacupunc- ture Parameters: limb tremor or minor tonic contraction of muscle, and put current controled in 4~7 mA; weak Electroacupuncture parameters: beard fretting or muscle mild beating, and the current controled in 1 ~3 mA. Then sampling after 24 hours. TNFα and IL-1βcontents of rat serum were measured by enzyme linked immunosorbent assay (ELISA) and did the statistical analysis. Results Compared with the normal group and sham operation group, the contents of TNFα adn IL-1β in model compare group were signifi cantly difference (P 〈0.01) ; compared with the model compare group, the contents of TNFα and IL-1β were significantly decrease in the electroacupuncture group (P〈 0.05~0.01); Compared with the weaker electroacupuncture group, the regulation of TNFα adn IL-1β in the stronger electroacupuncture group was significant (P〈0.05). Conclusion The therapeutic of different intensity electroacupuncture in the regula tion of inflammatory cytokines TNFα and IL-1β have differences in the rats of acute cerebral ischemia reperfusion. The therapeutic of strong stimulation is better than the weak stimulation.
