摘要
从草莓杂交育种的后代群体中选择出优系妙紫,以其茎段和茎尖为外植体,研究了侧芽和茎尖诱导培养及增殖快繁、生根的适宜培养基,以建立其组培快繁的技术体系。结果发现:(1)茎段和茎尖培养的适宜培养基是MS+0.5 mg/L BA+0.1 mg/L NAA,芽都能诱导萌发,且新梢生长健壮。(2)适宜的增殖快繁培养基是MS+0.5 mg/L BA+0.05 mg/L NAA,增殖系数较高,可达10倍,增殖方式是以腋芽增殖为主,新梢生长健壮。在激素水平增加的MS+1.0 mg/L BA+0.1 mg/L NAA培养基上,虽然增殖系数可达20倍,但芽丛长势弱。(3)适宜的生根培养基是1/2MS+0.5 mg/L NAA,新梢平均生根数达4.5条。
The superior strawberry line Miaozi was selected from the crossbreeding. In order to establish its tissue culture and rapid propagation system, its stalk section and shoot tip were used as explants to screen the suitable media for bud induction, proliferation and footage. The results were as follows. (1) The suitable medium for stalk section and shoot tip culture was MS + 0.5 mg/L BA + 0.1 mg/L NAA, in which the buds could germinate into the vigorous shoots. (2) The suitable medium for multiplication culture was MS + 0.5 mg/L BA + O. 05 mg/L NAA with the multiplication muhiple as i0 and sprouting in the form of axillary bud, and the shoots grew vigorously. Meanwhile, on the medium of MS + 1.0 mg/L BA + 0. 1 mg/L NAA, the multiplication multiple was 20, but the new shoots grew weaker. (3) The suitable rooting medium was 1/2MS + 0.5 mg/L NAA, in which the average root number was 4.5.
