摘要
①目的探讨硅元素在矽肺模型大鼠各器官组织中的分布情况。②方法雄性Wistar大鼠按体质量随机分为2组,即对照组、染尘组。染尘组依据染尘时间(1、3、5、7、14、21和28d)分为7个亚组,每组5只大鼠。取大鼠心、肝、脑、脾、肺、肾等器官组织,采用电感耦合等离子体质谱仪(ICP-MS)测定样品中硅元素的水平,并进行准确度和精确度实验;取每只大鼠右下肺最大横径处肺组织用于石蜡包埋切片,常规行HE染色进行病理学观察。③结果染矽尘大鼠各器官组织中硅元素水平在SiO2进入体内后即可上升,并随时间的推移呈升高趋势且分布存在差异。④结论矽肺模型大鼠各器官组织硅元素水平浓度分布大小依次为肺、心、肝、脾、肾、脑。ICP-MS法可以较好地检测硅元素,具有一定的意义及价值。
Objective The aim of the study is to probe the distribution of silicon levels in different organs in silicosis model rats by ICP - MS. Methods Rats were randomly divided into control group and 7 experimental groups according to different time (1,3,5,7,14,21and28d). Each group has 5 rats. Rats were sacrificed and their organ tissue samples such as heart,lung,liver, brain ,spleen and kidney were taken. The silicon levels were determined by inductively coupled plasma mass spectrometry( ICP - MS) ,in the same time,the degree of accuracy and precision were tested. Moreover, Lung tissue were stained by haematoxylin and eosin(HE) ,and observed by a microscope. Results The silicon levels in different organs of silicosis - model - rats raised right after SiO2 enter the body, stepped up as the time went on and distributed areas were different. Conclusion The concentration of silicon in silicosis -model -rats organ tissues descents in sequence,the order is lung,heart,liver,spleen,kidney and brain. ICP- MS is a better way to detect silicon in rats. It's indicated that there exist some value and meaning in detection silicon levels by ICP - MS.
