摘要
目的:探讨曲美他嗪对糖尿病心肌病大鼠心肌细胞纤维化的影响。方法:取Sprague-Dawley雄性大鼠30只,随机分为糖尿病心肌病组(D组,n=10),糖尿病心肌病曲美他嗪干预组(T组,n=10),另取健康大鼠10只,作为正常对照组(N组,n=10)。T组给予曲美他嗪[7.5mg/(kg.d)]灌胃,D组和N组给予等量蒸馏水灌胃,每日2次,共8周,每周称重调节药物用量。观察并比较3组试验动物心肌细胞纤维化的相关指标。结果:D组心肌羟脯氨酸水平(Hyp)(4.8±2.6)mg/g明显高于N组(2.5±1.0)mg/g和T组(2.7±1.7)mg/g,P均<0.05,且T组明显低于D组(P<0.05)。D组心肌间质胶原容积百分数(ICVF,5.6±2.1)明显高于N组(2.6±1.3)和T组(4.0±1.5),P均<0.05,且T组明显低于D组(P<0.05)。结论:曲美他嗪对抑制糖尿病心肌病大鼠心肌细胞纤维化有一定的作用。
Objective: To investigate the effects of trimetazidine on cardiac fibrosis in rats with experimental diabetic cardiomyopathy(DC).Methods: DC was induced in male Sprague-Dawley rats by STZ intraperitoneally injection.Rats randomly received saline(n=10) or trimetazidine(7.5 mg per kg weight per day,n=10) for 8 weeks by gavage.Another 10 healthy rats were served as controls.Myocardial hydroxyproline content as well as interstitial collagen volume fraction(ICVF) were determined.Results: Myocardial hydroxyproline and ICVF were more significantly reduced in trimetazidine treated DC rats as compared to saline treated DC rats(all P0.05).Conclusion: Trimetazidine could decrease the concentration of myocardial hydroxyproline and ICVF in diabetic cardiomyopathy rats.
