人乳腺癌及癌旁组织磷酸果糖激酶1及糖酵解水平的检测 被引量:1

Expression of Phosphofructokinase-1 and Glycolytic Level in Human Breast Cancer and Paracancer Tissues

摘要 目的:比较人乳腺癌及癌旁组织中糖酵解的水平差异,并探讨磷酸果糖激酶1的表达对糖酵解水平的影响。方法:采集3种不同临床分期共33对人乳腺癌及癌旁组织。酶法分析组织中乳酸含量、乳酸脱氢酶活力、己糖激酶活力、磷酸果糖激酶-1活力和丙酮酸激酶活力,用来反映糖酵解的水平;采用Western blot技术检测组织中磷酸果糖激酶-1的表达。结果:癌组织的糖酵解水平和磷酸果糖激酶1表达量均高于癌旁组织,而且,晚期癌组织的糖酵解和磷酸果糖激酶1表达量水平高于早期乳腺癌组织。结论:在人乳腺癌的发生发展过程中,糖酵解水平的增加可能与磷酸果糖激酶1的表达量增加有关。 Objective: To compare the glycolysis rate in human breast cancer and paracancer tissues,and detect the effect of phosphofructokinase-1 expression in glycolysis rate.Methods: Thirty-three couples of human breast cancer and paracancer tissues in three different clinical stages were collected.Lactic acid content,lactate dehydrogenase activity,hexokinase activity,phosphofructokinase-1 activity and pyruvate kinase activity,which reflect glycolysis status,were measured by enzymatic methods;the phosphofructokinase-1 expression was detected using Western blot analysis in both human breast cancer and paracancer tissues.Results: The glycolysis rate and total phosphofructokinase-1 expression increased in human breast cancer tissues and superior clinical stages.Conclusion: The enhancement of glycolytic activity may due to high expression of phosphofructokinase-1 in human breast cancer.
