摘要
目的探讨广西红水河流域壮族长寿家系微粒体三酰甘油转移蛋白(microsomal triglyceride transferprotein,MTP)-493G/T基因多态性与血脂水平的相关性。方法应用PCR-RFLP方法对广西红水河流域壮族长寿家系(HZL组)MTP基因的-493G/T位点进行基因分型,并与当地非壮族长寿家系(HNL组)、壮族非长寿家系(HZN组)进行比较;测定3组血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇水平,分析几种基因型与血脂水平的关系及其在长寿中的作用。结果 3组间等位基因及基因型频率差异无统计学意义(P>0.05);HZL组和HNL组TT基因型个体低密度脂蛋白胆固醇水平及HZL组TT基因型个体高密度脂蛋白胆固醇水平均明显低于GG和GT基因型个体(P<0.05);HZN组内各基因型个体血脂水平比较差异无统计学意义(P>0.05)。结论广西红水河流域壮族长寿人群存在MTP-493G/T单碱基突变,TT基因型个体与低密度脂蛋白胆固醇水平降低有关。
Objective To investigate the relationship between the microsomal triglyceride transfer protein(MTP)-493G/T gene polymorphism and the level of serum lipids in Zhuang longevous families in the Red River Valley of Guangxi.Methods The genotype and allele of MTP-493G/T were determined with PCR-RFLP in Zhuang longevous families in the Red River Valley of Guangxi(HZL group).The results were compared with Non-Zhuang longevous families(HNL group) and Zhuang non-longevous families(HZN group) in the Red River Valley of Guangxi.The serum levels of total cholesterol,triacylglycerol,low density lipoprotein-cholesterol(LDL-C) and high density lipoprotein-cholesterol(HDL-C) were determined in all subjects.The relationship between genotype of MTP-493G/T and serum lipids and their role in longevity were analyzed.Results There were no significant differences in genotype and allele frequencies among the three groups(P0.05).The TT genotype individuals had lower level of LDL-C than the GG or GT genotype in HZL and HNL group,and had lower levels of HDL-C in HZL group(P0.05).There were no significant differences in the serum lipid levels among the three genotypes in HZN group(P0.05).Conclusion MTP-493G/T gene mutation is found in Zhuang longevous population in the Red River Valley and the TT genotype is closely correlated with the decrease of LDL-C level.
