摘要 背景:骨髓间充质干细胞移植影响大鼠肝脏癌变过程中肝脏血管报道目前不多。目的:观察骨髓间充质干细胞移植对大鼠肝脏癌变过程中血管生成的影响。方法:将30只Wistar大鼠随机摸球法均分成骨髓间充质干细胞移植组、单纯造模组和空白对照组,前2组以二乙基亚硝胺为诱癌剂制备肝癌模型,空白对照组仅给予正常食水作对照。结果与结论:单纯造模组8只大鼠形成肝癌,骨髓间充质干细胞移植组9只大鼠形成肝癌,肝脏携带SrY的阳性细胞,MRI影像中可见较多占位性病变,而单纯造模组占位病变数量明显较少,两组比较差异有显著性意义(t=2.45,P=0.026);骨髓间充质干细胞移植组大鼠血管内皮细胞生长因子和微血管密度平均阳性表达率均高于单纯造模组(P<0.05)。说明在二乙基亚硝胺制备的大鼠肝癌模型中,骨髓间充质干细胞可通过促进血管的生成而促进肝癌的生长。 BACKGROUND:There are few reports about the effects of bone marrow mesenchymal stem cells(BMSCs) transplantation on vascularization during hepatocarcinogenesis in rats.OBJECTIVE:To observe the effects of BMSCs transplantation on vascularization during hepatocarcinogenesis in rats.METHODS:Thirty Wistar rats were divided into BMSCs transplantation group,pure model group and blank control group randomly.The first two groups were treated with diethylnitrosamine to prepare hepatoma models.The blank control group was treated with water as control.RESULTS AND CONCLUSION:Eight rats in the pure model group and nine rats in the BMSCs transplantation group were proved to be with liver cancer,and the SrY + cells were seen in the BMSCs transplantation group.MRI showed that there were more space-occupying lesions in the liver of the BMSCs transplantation group compared with the pure model group(t = 2.45,P = 0.026).The average positive expression rate of vascular endothelial growth factor and microvessel density in the BMSCs transplantation group were higher than those in the pure model group(P 0.05).In the hepatoma models of rats prepared with diethylnitrosamine,BMSCs can promote the vascularization resulting in the development of liver cancer.
