摘要
目的 观察益骨胶囊含药血清在成骨.破骨细胞共育体系中对SD大鼠成骨细胞表达骨保护素(OPG)mRNA的影响。方法 取1d龄SD大鼠颅骨分离培养成骨细胞(0B);取5d龄SD大鼠四肢长骨分离培养破骨细胞(OC)。建立上清相通但细胞间不相互混杂的平面式成骨一破骨细胞共育体系,实验分为含药血清组和对照组。将10月龄SD雌性大鼠分为益骨胶囊灌胃组和生理盐水对照组,制备含药血清和对照血清。检测共育体系中成骨细胞OPG mRNA的表达。结果含药血清组与对照组比较,OPG mRNA表达明显升高(8.2567±0.1118比3.3350±0.9854),差异有统计学意义(P〈0.01)。结论益骨胶囊含药血清在共育体系中通过刺激OB表达OPG来实现对OC的活性和凋亡的调节。
Objective To investigate the impact of Yigu capsule- containing serum on the expression of osteoprotegerin (OPG)mRNA in SD rat osteoblasts in an osteoblast and osteoclast co-culture system. Methods Osteoblasts (OBs) and osteoclasts (OCs) were isolated and cultured from the skulls of 1-day-old and the fetal long bones of 5-day-old SD rats for establishment of the serum interlinked but intercellular unmixed plane OB and OC co-culture system. The rats were then divided into medicine-containing serum and control groups. Furthermore, the 10-month-old female SD rats were divided into Yigu capsule-gastric lavage and NS groups for preparations of medicine containing serum and control serum. FQ-PCR method was used to detect the OPG mRNA expression in OBs in the co-culture system. Results Compared with the control group, the medicine containing serum group had a significant increase in OPG mRNA expression (8.2567±0.1118 vs 3.3350±0.9854, P〈0.01). Conclusion In the co-culture system, Yigu capsule containing serum may regulate the activity and apoptosis of OC by stimulating OB to express OPG.
