摘要
在pH值为8.0的Britton-Robinson缓冲介质中,盐酸吡格列酮与溴甲酚绿反应,形成离子缔合物,最大褪色波长位于616 nm处,同时在450 nm处出现增色效应,在最大褪色与最大增色波长处,盐酸吡格列酮的浓度分别在2.81×10 6~1.52×10 5mol/L和7.70×10 6~1.54×10 5mol/L范围内遵守比尔定律,表观摩尔吸光系数ε分别为6.08×104和1.56×104L/(mol cm),检测限分别为1.99×10 7和2.51×10 7mol/L.若用双波长叠加,检测限更低.方法用于市售药品中盐酸吡格列酮含量的测定,所得结果满意.以血浆和尿液样品为基体加入标准溶液作回收率试验,测定回收率在96.7%~103.6%之间,测定值的相对标准偏差均小于3.70%.
In Britton-Robinson buffer medium of pH 8.0,bromocresol green reacts with pioglitazone hydrolchloride,forming ion-association complexes,maximum fading wavelength of complexes is 616 nm,the maximum absorb wavelength locates 450 nm,the concentration of pioglitazone hydrolchloride follows Beer’s law for the tight absorption in the different range of 2.81×10 6~ 1.52×10 5mol/L and 7.70×10 6~1.54×10 5mol/L respectively,the apparent molar absorptivities are 6.08×104 L/(mol cm) and 1.56×104 L/(mol cm) and the detection limits are 1.99×10 7mol/L and 2.51×10 7mol/L respectively.If the determination process is dealt with double wavelength,Lower the detection limits.Samples of tablets and capsules of pioglitazone hydrochloride were analyzed by the proposed method,and the amounts of PGH found were checked quite well with the labelled values,the results are satisfactory.Recovery and precision of the method were tested by the standard addition method using samples of human blood serum and urine as matrixes,values of recovery found were in the range of 96.7% and 103.6%,and values of RSD found were less than 3.70%.
