悉生豚鼠体内复方双歧杆菌制剂对福氏志贺氏菌的作用 被引量:4

摘要 复方双歧杆菌制剂中,双歧杆菌(Bif.bifidum),乳酸杆菌(L.acidophilus),粪链球菌(Str.faecalis)三种菌都是人和动物肠道正常菌群之一,要研究它们和肠道致病菌之间的关系,在普通动物体内是无法观察到.在国内我们首次应用悉生豚鼠进行复方双歧杆菌制剂对福氏志贺氏菌作用的研究,发现在悉生豚鼠体内复方双歧杆菌制剂对感染后的豚鼠有较明显的保护作用,保护率为75%,实验组未获得保护的豚鼠死亡时间较对照组延长6~10天,而对照组豚鼠在感染后24~48小时全部死亡,解剖后在肠道分泌物中分离出福氏志贺氏菌.本实验证明:复方双歧杆菌制剂是治疗腹泻较好的生物制剂,尤其对因菌群失调而引起的腹泻更是理想的治疗方法。 Included in the compound preparations of Bifidobacterium are Bifido-bacterium bifidum,Lactobacillus acidophilus &.Strep- tococcus faecalis,all being constituents of normal microbial flora of the intestine of human beings.In order to find out whether there is any antimicrobial action of the preparation against enteropathogenie microbes in vivo,gnotobiotic guinea-pigs were uti- lized as an experimental animal model which was found to be suitable,and as a host,its sensitivity was comparable to that of human beings. Cermfree guinea-pigs delivered form hea- lthy full-term pregnanted animals by caesar- cart section and reared aseptically in isola- tors with microbial surveillance at intervals. On the 3rd day after delivery,100,000 live bacteria/per capita of Strep.faecalis were given by mouth.On the 23rd day,the ani- mals were infected with a virulent.Strain of Shigella flexneri 2a,a dose of 100,000 viable bacteria/per capita in 0.2 ml by mouth.On the 2nd day of infection with the enteric pathogen,the compound bacterial preparations of Bifidobacterium,Lactobacillus & Strepto. faecalis(100,000 microbes per species each of the above-mentioned microbes)/10mg./capita by oral ruote.This dose of medicine was continued for 4 successive days,with one dose/day.Observations of the colonization of the bacterial flora including the introdu- ced pathogenic microorganism were made at 2-4 days' intervals.The experimental results indicated that the introduced three spp.nor- mal bacterial flora in the preparation exhi- bited marked protection of the animal host with a protective rate of 75%,and a longer survival period,(6-10 days)in those succu- mbed,as compared with the controls which were found died of infection of Shigellosis within 24~48 hours,With a mortality rate of 100%.On autopsy,from the intestinal contents of those animals died of the infec- tion Shi.flexneri were found in pure cultures. The experiments indicated that the com- pound viable preparations of Bifidobacterium etc.was an effective therapeutic micro-eco- logical agent for infectious diarrhoae Bifido- bacterium experimental Shigellosis in gnoto- biotic guinea-pigs.It seems to be an ideal methed for modulation for disturbed state of intestinal microbial requillibrum during diarrhoae.
