学习记忆过程中海马S100β和NOS表达的变化及其相关性 被引量:10

摘要 目的 探讨大鼠学习记忆训练过程中海马 S10 0 β和一氧化氮合酶 (NOS)表达的变化及其相关性。 方法  Y型电迷宫和跳台反射仪连续训练一周作为空间学习记忆的训练组 ,随机操作为假训练组 ,而自由活动作为正常对照 ;免疫组化显示 S10 0 β、n NOS和 i NOS的表达情况并在高倍镜下进行阳性细胞计数及方差分析。 结果 训练组大鼠海马及齿状回S10 0 β和 n NOS均呈高表达 ,阳性细胞数显著多于假训练组和对照组 (P <0 .0 1) ;训练组尚有少量 i NOS表达 ,其分布与S10 0 β+星形胶质细胞一致。 结论  S10 0 β和 NOS的表达上调在学习记忆机制中具有重要作用 ,S10 0 β和一氧化氮 (NO)之间的协同作用可能与长时记忆的获得有关。 Objective To obseve the expressions of S100β and nitric oxide synthase (NOS) in hippocampus and dentate gyrus during training of learning and memory and to explore the role of S100β and NOS in the mechanism of learning and memory Methods Ten rats (training group) received one week regulation training with Y maze and stair jumping instrument The sham training group (n=8) rats were trained randomly in Y maze and stair jumping instrument and the control group rats moved freely at equal times Immunohistochemical assay was used to investigate the expressions of S100β、nNOS and iNOS S100β + and nNOS + cells were counted at a magnification of 400×, in four microscopic fields and assessed for statistical significance with One Way ANOVA and LSD Results Training group rats had increased numbers of S100β + astrocytes and nNOS + neurons in hippocampus and dentate gyrus compared with sham training group and control group rats (P<0 01) Furthermore,in the training group,but not in the sham training group and control group, rats also show a few iNOS + astrocytes and its distribution in hippocampus and dentate gyrus correlated closely with the expression levels of S100β Conclusion The training related increases in S100β and NOS expression are important to the mechanism of learning and memory Meanwhile,the synergism between S100β and nitric oxide (NO) might have relevance to acquisition of long term memory However,the exact regulation mechanism needs further investigation
