摘要
本试验就蜡状芽孢杆菌大罐深层培养法进行了初步研究,基本解决了蜡状芽孢杆菌大罐液体深层培养条件下芽孢生成、适宜培养基的选择、培养方法的建立以及菌体的收集方法等技术问题,大罐深层培养生产工艺的建立较固体培养法产量大为增加,污染耗损降至最低,减少劳动强度和繁琐的操作,而且产品质量得到保证和提高.
Cujunshen(Cereobiogen)is a preparation of live Bacillus cereus used for treatment of diarrheas both in human beings,and in poultry and livestocks.For mass production, a fermentor of 1500 l in capacity charged with 1000 l of a medium composed of 50 per cent of acid hydrolysate of casein,pH 6.8,is used for cultivation.After inoculation with the seed culture DM 432,the culture fluid is incubated at 32-34℃.for 36-48 hr under con- tinuous aeration and agitation(280 r.p.m.). Under these conditions,90-99 -per cent of cells will form spores.Thereafter,the bacter- ial mass is collected by continuous centrifu- gation at 15000 r.p.m.The wet mass is mixed with CaCo_3 in a proportion of 1:2. The lump is spread on a glass plate and dried at 60-70℃.in an oven.The powder is ground,sifted through a 80-mesh sieve,and compressed into molded tablets. The use of fermentor cultivation has several advantages.In comparison with the method of plate cultivation,the productivity increases considerably with minimum costs of operation and equipraents,including manpo- wer,less losses due to contamination.Furt- hermore,strenuous labor and tedious proced- ures of processing can be avoided,and the quality of the products can be assured.
