摘要
选取45个微卫星位点,对爆发马立克氏病的4只患鸡的淋巴细胞和正常的组织及3号鸡的肿瘤组织,提取核酸,利用45个所选微卫星引物进行PCR扩增,变性聚丙烯酰胺凝胶电泳检测微卫星不稳定性的变化。研究结果表明,在4只患鸡中有39个微卫星位点发生微卫星不稳定性的变化,其中3号鸡发生变化概率最高,证实同个体的淋巴细胞和肿瘤组织同位点发生MSI变化的符合率达到75.6%。
Forty-five microsatellite sites were selected,nucleic acid was extracted from the lymphocyte and normal tissue of 4 sick chickens with Marek's disease as well as the tumor tissue of chicken No.3,PCR amplifications were undertaken by using 45 special primers,and then the denatured polyacrylamide gel electrophoresis was used to analyze the changes in the instability of microsatellite.The results showed that 39 microsatellite sites generated microsatellite instability among 4 sick chickens,and the probability of microsatellite instability in chicken No.3 was maximum.It was confirmed that the coincident rate of MSI change at the same site between lymphocyte and tumor tissue from the identical individual reached 75.6%.
