PI3K/Akt/mTOR信号通路及mTOR抑制剂在泌尿系统肿瘤中的研究进展 被引量:11

摘要 PI3K/Akt/mTOR信号通路在血管生成、蛋白合成以及细胞生长代谢中发挥着关键作用,在多种实体性恶性肿瘤中已发现该信号通路调节异常。随着PI3K/Akt/mTOR信号通路相关抑制药物的相继问世,尤其是在治疗晚期肾癌中显示出一定的临床疗效,对该通路以及相应抑制药物的深入研究,特别是在泌尿系统肿瘤的应用,成为热点之一。该文总结了PI3K/Akt/mTOR信号通路的发生机制及其在肿瘤形成过程中的作用,并就mTOR抑制剂在泌尿系统肿瘤中的研究进展进行综述。 PI3K/Akt/mTOR signaling pathway plays a vital role in the angiogenesis,protein synthesis,cell growth and metabolism,and dysregulation of this pathway has been found in many substantive malignant tumors.In recent years,the inhibitors of PI3K/Akt/mTOR signaling pathway have been developed in succession,which have exhibited definite clinical effects on advanced renal cell carcinoma.As a result,further research on this pathway and its inhibitors,particularly their applications in urologic malignancies,has become one of the hot research areas.This paper focuses on the mechanism of PI3K/Akt/mTOR signaling pathway and its roles in the development of tumors,and the research progress of mTOR inhibitors in urologic malignancies is also reviewed.
