摘要
目的探讨甲醛暴露人群对机体内源性小分子代谢产物的影响。方法借助超高效液相色谱与串联四级杆飞行时间质谱(UPLC-Q-TOF/MS)检测技术,对甲醛暴露组与对照组的尿液样品进行检测,建立尿液代谢轮廓图,并进一步借助非监督模式的主成分分析(PCA),从体内微观角度证明了甲醛对机体整体代谢网络的干扰。结果建立了甲醛职业接触者尿中内源性代谢产物的UPLC-Q-TOF/MS测定方法,确定甲醛职业接触与未接触者尿液代谢轮廓图的差异;明确了表征甲醛对机体损伤的内源性生物标志物。结论从生物体内整体代谢方面说明了甲醛对职业接触者的毒性危害。
Objective To investigate effects of formaldehyde on endogenous metabolites of urine. Methods The urinary samples of the formaldehyde eontact group and control group were analyzed by UPLC -Q-TOF/MS, combined with principal component analysis(PCA). The overall disease pattern of the metabolic network was established based on metabolic profiles in different groups. Results The different trajectory of urine was visually noted from metabolic profiles between 2 groups. Metabolomics appeared to be suitable for toxicity studies of formaldehyde, and a biomarker structure was identified. Conclusion The science connotation of the toxicity of formaldehyde may be elucidated by Metabonomics, according to metabolites variance in certain disease pattern organism.
