缺氧诱导因子1α和胰岛素在胰腺癌中的表达及相关性 被引量:3

Expression of hypoxia inducible factor-1a and insulin in pancreatic cancer and their correlation

摘要 目的 探讨缺氧诱导因子1a(HIF-1a)蛋白和胰岛素在胰腺癌组织中的表达及其相关性。方法采用免疫组织化学sP法检测65例胰腺癌中心癌组织和边缘癌组织中HIF-1a和胰岛素的表达情况;Westernblot法检测28例胰腺癌中心癌组织和边缘癌组织中HIF-1a蛋白和胰岛素的表达水平,并分析其相关性。结果HIF-1a蛋白在65例胰腺癌中心癌组织和边缘癌组织中的阳性表达率分别为70.8%和66.2%,差异无统计学意义(P〉0.05)。HIF—1a蛋白在28例胰腺癌中心癌组织和边缘癌组织中的表达量分别为0.338±0.030和0.327±0.035,差异无统计学意义(P〉0.05)。胰岛素在胰腺癌中心癌组织和边缘癌组织中的阳性表达率分别为41.5%和80.0%,差异有统计学意义(P〈0.05)。胰岛素在28例胰腺癌中心癌组织和边缘癌组织中的表达量分别为0.191±0.016和0.584±0.022,差异有统计学意义(P〈0.05)。相关性分析显示,胰腺癌边缘癌组织中HIF-1a蛋白与胰岛素水平有关(r=0.374,P〈0.05),呈明显正相关性(r=0.89,P〈0.05);胰腺癌中心癌组织中HIF.10t蛋白与胰岛素水平无相关性(r=-0.145,P〉0.05)。结论通过调控HIF-1a蛋白的表达,胰岛素可增强胰腺癌细胞的侵袭与转移能力。 Objective To explore the correlation between the expressions of hypoxia inducible factor-let (HIF-let) and insulin in pancreatic cancer. Methods HIF-1a and insulin expression was detected by immunohistochemistry in the center and the edge of pancreatic adenocarcinoma specimens of 65 cases. Western blot was used to detect HIF-1a expression and insulin level in the center and the edge of pancreatic adenocareinoma specimens of 28 cases. The relationship between HIF-1a expression and insulin level in the pancreatic cancer was analyzed. Results The results of immunohistochemistry and Western blot showed that HIF-1a protein expression was high in both the center and the edge of pancreatic cancers ( P 〉 0.05) , and insulin level was significantly higher at the edge of specimen than that in the center (P 〈 0. 05 ). HIF-1a protein and insulin levels were positively correlated at the edge of cancer tissue (r = 0. 374, P 〈 0.05 ) , but no significant correlation between them in the center of cancerous tissue ( r = - 0. 145, P 〉 0. 05). Conclusion Insulin may promote the local invasion and metastasis of pancreatic cancer by upregulating HIF-1 a.
