姜黄素抗辐射诱导内皮细胞凋亡信号的转导机制 被引量:1

摘要 目的探讨姜黄素对辐射诱导血管内皮损伤保护作用及其信号转导机制。方法对人脐静脉内皮细胞(HUVEC)分离培养后分组以直线加速器进行6Gy8线照射,并设未照射组为对照。分别通过光镜及电子显微镜观察细胞形态和超微结构改变;流式细胞仪法观察各组的凋亡率和坏死率;Westernblot法检测各组ERK1/2、p-ERK1/2表达;给以MEK1/2通路的特异性阻断剂U0126处理后检测各组细胞凋亡情况。结果细胞经照射后,形态及超微结构改变均显示为典型的凋亡改变,而姜黄素预处理组的细胞调亡率和坏死率均明显低于其他实验组,差异有统计学意义(P〈0.05)。姜黄素组ERK1/2、p-ERK1/2表达高于对照组,加入U0126后HUVEC凋亡率明显升高,差异有统计学意义(P〈0.05)。结论放射线照射可导致HUVEC凋亡和坏死,姜黄素对放射后HUVEC凋亡和坏死有保护作用,并且这种保护作用与MEK1/2信号通路激活有关。
