养精胶囊对Leydig细胞增殖、凋亡和睾酮合成的影响 被引量:3

Effects of Yangjing Capsules on proliferation, apoptosis and testosterone synthesis of Leydig cells

摘要 目的探讨养精胶囊对Leydig细胞增殖、凋亡和睾酮合成功能的影响。方法在Leydig中加入不同剂量养精胶囊提取液24h、48h、72h后,用MTT法检测各组的吸光值;用流式细胞仪分析Leydig细胞在加入不同剂量养精胶囊提取液48h后的细胞凋亡率;用化学发光法测定Leydig细胞在加入不同剂量养精胶囊提取液24h后合成T的变化。结果Leydig细胞在加入中、高剂量养精胶囊提取液48h和72h后,细胞增殖明显加快,其吸光值与空白组相比差异有统计学意义(P〈0.05);在Leydig细胞中加入中、高剂量养精胶囊提取液48h后,细胞凋亡率和死细胞率明显降低,活细胞率明显提高,与空白组相比差异有统计学意义(P〈0.01);在Leydig细胞中加入中剂量养精胶囊提取液24h后,睾酮合成明显增加,与空白组相比差异有统计学意义(P〈0.01)。结论养精胶囊可能通过影响Leydig细胞的增殖、凋亡,增强其分泌功能等多途径,而提高睾酮合成的功能,并与时间和剂量有关。这也证实了益肾填精法能直接作用于Leydig细胞而影响睾酮的合成。 Objective To investigate the effects of Yangjing Capsules on proliferation, apoptosis and testosterone synthesis of the Leydig cells. Methods Proliferated Leydig cells treated with different doses of Yangjing Capsule extract for 24h, 48h and 72h were respectively detecetd by MTT. The apoptosis rate of the Leydig cells was measured by flow cytometry treated with different doses of the extract for 48h. Concentration of T was assayed by chemiluminescence after the Leydig cells were treated with different doses of the extract for 24h. Results Prolifera- tion of the Leydig cells increased significantly when they were treated with medium and high doses of the extract for 48h and 72h, compared with that of the control group (P〈0.05). After the Leydig cells were treated with medium and high dose of the extract for 48h, the apoptosis cells and dead cells significantly decreased, whereas the living cell rate significantly increased, compared with those of the control group (P〈0.01). The synthesis of T in the Leydig cells was increased significantly when they were treated with the medium dose extract for 24h compared with that of the control group (P〈0.01). Conclusion Yangjing Capsules might improve the function ofT synthesis by promoting Leydig cells 'proliferation, reducing its apoptosis and, enhancing its secretion function. It is related to the timing and dosage. It also suggests that reinforcing kidney essence can effect on the Leydig cells and promote testosterone synthesis.
