显微拉曼光谱法定量检测碳氧血红蛋白饱和度 被引量:5

Quantitative detection of carboxyhaemoglobin saturation by microscopic Raman spectroscopy

摘要 采用显微拉曼光谱技术对不同饱和度碳氧血红蛋白(HbCO)进行测定,通过偏最小二乘法(PLS)和间隔偏最小二乘法(IPLS)建立模型,得出线性回归方程.结果表明,经过筛选变量信息所建立的间隔偏最小二乘模型(IPLS),当血红蛋白系列样品中碳氧血红蛋白含量为0~100%时,线性相关系数的平方为0.99,检测限为5.37%,完全能满足临床和法医学上CO中毒程度的检测.此方法具有快速、简便、直接、对样品无损等优点,有望成为检测碳氧血红蛋白饱和度的新方法. To explore a new method for detect the carbon monoxide poisoning degree and carboxyhaemoglobin saturation in body,micro-Raman spectroscopy in combination with partial least squares model and interval partial least squares model was used for investigate the different degree of saturation of carboxyhaemoglobin.Multiple statistical analysis result shows that when the degree detection range of saturation of carboxyhaemoglobin is from 0 to 100%,the interval partial least squares model after screening variable information obtained the correlation coefficient was 0.99,and the detection limit of saturation is 5.37%,which could meet CO poisoning degree detection on clinical and forensic science.This technique using Raman spectroscopy is quick,convenient and nondestructive,and may potentially become a new method for direct measurement of degree of saturation of carboxyhaemoglobin.
