摘要
为了探讨鸡新城疫病毒(Newscastle disease virus,NDV)感染不同细胞的病变特性,选用NDV强、弱毒株感染鸡胚成纤维细胞(DF-1)、仓鼠肾细胞(BHK-21)和人宫颈癌细胞(HeLa).结果表明:NDV强毒株在DF-1、BHK-21、HeLa上均能增殖,在DF-1、BHK-21、HeLa上产生的细胞病变(cytopathic effect,CPE)以发生细胞融合和形成合胞体为主要特征,在DF-1上增殖速度较快,HA效价也高;NDV弱毒株含10μg/mL胰蛋白酶的DMEM培养基上,DF-1、BHK-21、HeLa可引起细胞变圆、脱落,没有典型的CPE,盲传15代CPE特征未发生改变,血球凝集(HA)效价较低.可见,NDV在其种属来源相同的DF-1中更适宜增殖,DF-1可用作NDV感染的最佳细胞模型.
Virulent and attenuated newscastle disease virus(NDV) were adapted to grow in the DF-1,BHK-21 and HeLa cell lines in order to investigate different cytopathic effect(CPE) in various cell lines.The results showed that virulent NDV could produce typical cytopathic effect after serial passages in various cell lines,as indicated by CPE of rounding,degeneration and the formation of synctia in virous cell line.But the HA titer and proliferation rate were better in DF-1 cell line than those in BHK-21 and HeLa cell lines.When the concentration of trypsin was 10μg/mL in DMEM,attenuated NDV was confirmed circular and desquamated without typical CPE,no modification in the CPE properties was detected after fifteen passages,also the HA titer displayed zero.It appears that NDV strain grew in DF-1 culture has retained its biological homology,so the use of DF-1 cell line could be the best cell culture for the infection of NDV.
