Expression and Significance of Pleiotropin in Periodontitis

摘要 目的:研究多效生长因子(pleiotropin,PTN)在牙周炎中的表达及意义。方法:应用免疫组织化学、West-ern免疫印迹方法,对雄性Wistar大鼠50只分为炎症组、恢复组和对照组分别检测PTN蛋白表达的情况。结果:PTN的阳性产物位于龈上皮、沟内上皮和结合上皮细胞胞浆中,呈棕黄色颗粒偶见胞核中亦有表达。炎症组中MK的表达主要集中于棘细胞层和颗粒细胞层,表达量为弱阳性和中强阳性。恢复组中PTN的表达主要集中于棘细胞层和基底细胞层,表达量为强阳性和中强阳性。对照组中PTN表达较少。Western免疫印迹显示PTN在炎症组和恢复组中表达有统计学意义(P<0.01)。Western免疫印迹与免疫组织化学检测结果基本一致(P<0.05)。结论:PTN同时具有致炎和促进组织修复的功能,但促进组织修复的作用更明显。 Objective: To study the expression and significance of midkine in periodontitis. Methods: Immunohis- tochemistry and Western-blotting analysis for PTN were performed on 50 male Wistar rats which wered divided in- to inflammation group, recovered group and control group. Results: The positive products (brown-yellow grains) of PTN lled in the cytoplasm of gingival epithelium, gingival sulcus epithelium and junctional epithelium cells and occasionally in the nucleus of cells. Positive products expressed major in the spinous cell layer and granular cell layer in the inflammation group, and the expressions were weak-strong and mid-strong positive. Positive products ex- pressed major in the spinous cell layer and basilar cell layer in the recovered group, and the expressions were mid-strong and strong positive. The expression was less in the control group. Western-blotting half-quantity analy-sis: there were obvious statistical significance between the inflammation group and the recovered group (P〈0.01). The result of immunohistochemistry was the same to the result of Western-blotting (P〈0.05). Conelusion: PTN has the functions of bringing about inflammation and promoting the injured tissue to recover. The function of recov- ery was more remarkable.
