摘要
己二酸N-羟基琥珀酰亚胺酯(简称NHS-AA)已被证明用于胶原的交联改性具有低毒、反应活性高、生物相容性好的特点。为了进一步优化蛋白交联剂NHS-AA交联改性胶原的反应条件,对其水解动力学性质进行了研究。结果表明:在磷酸盐缓冲溶液中,NHS-AA的水解速率随溶液pH值的升高而加快,随温度的升高也呈增大趋势,平均温度效应系数为2.25,水解活化能为52.39kJ/mol,其水解反应初期符合一级动力学方程。
Adipic acid N - hydroxysuccinimide ester ( NHS - AA) was reported to be collagen cross - link with low toxicity, high reactivity and excellent biocompatibility. In order to optimize the reaction conditions of NHS - AA cross - linked with colla- gen, hydrolysis kinetic of NHS - AA was investigated. The results show that the hydrolytic rate of NHS - AA in the phosphate buffer solution increases with the pH and the temperature increases. The average temperature coefficient is 2.25 and the activation energy of hydrolysis is 52.39kJ/mol. The hydrolysis rate of NHS - AA could be characterized by first - order kinetics equation.
