骨水泥单体对心肌细胞凋亡的诱导及其对Bcl-2、Bax蛋白表达的影响 被引量:1

摘要 目的探讨骨水泥单体(MMA)致SD乳鼠心肌细胞凋亡作用及其发生机制。方法新生1-3dSD大鼠的心肌细胞分6组:组1为空白对照组;组2-6分别用0.5、1.0、2.0、4.0、8.0mg/mlMMA作用2h。CCK-8法测细胞存活率,TUNEL染色法测细胞凋亡指数,Western blot测Bcl-2、Bax蛋白表达。结果与组1相比,MMA作用心肌细胞2h后,细胞存活率随MMA浓度的增高而逐渐降低,组2-6心肌细胞凋亡指数呈浓度依赖性增高,同时抗凋亡蛋白Bcl-2表达降低,促凋亡蛋白Bax表达增高。结论 MMA可诱导心肌细胞凋亡。其机制可能与抑制Bcl-2表达和促进Bax表达有关。 Objective To investigate the effects of methyl methacrylate(MMA) on the apoptosis of cardiomyocyte from newborn SD rats and its mechanism. Methods Cardiomyocytes from newborn SD rats were prepared and divided into six groups.Group A was taken as blank control.Groups of B,C,D,E and F were treated with MMA of 0.5,1.0,2.0,4.0 and 8.0 mg/ml respectively for 2 hours.CCK-8 assay was used to detect the survival rate of cardiomyocytes,TUNEL cell staining was used to examine the apoptosis index,and the expressions of Bcl-2 and Bax were detected by Western blot. Results Compared to group A,after incubated with MMA for 2 hours,as the concentration of MMA increased in groups of B,C,D,E and F, the survival rate of cardiomyocytes was decreased,the apoptosis index was increased,the expression of Bcl-2 was decreased and the expression of Bax was increased. Conclusion MMA could induce the apoptosis of cardiomyocyte.The possible mechanism is related to the inhibition of the expression of Bcl-2 and the increase of the Bax expression.
