摘要
目的探讨人脂肪来源间充质干细胞(hAD-MSCs)在乳腺肿瘤微环境中对乳腺癌细胞的影响。方法采用Transwell体系间接共培养人乳腺癌细胞系(MCF-7)和hAD-MSCs,对共培养后的肿瘤细胞进行细胞形态,EMT相关标志以及肿瘤特性的检测。结果与对照组相比,共培养之后的MCF-7细胞发生上皮间质转化。与hAD-MSCs共培养能明显促进乳腺癌细胞的侵袭和迁移,但对细胞周期和增殖能力没有明确的影响。结论人脂肪来源间充质干细胞可诱导乳腺癌细胞上皮间质转化。
Objective To study the effect of human adipose tissue derived MSCs(hAD-MSCs) in breast tumor micro-environment.Methods Breast cancer cell line(MCF-7) was cultured alone or indirectly co-cultured with hAD-MSCs using Transwell system.The co-cultured MCF-7 cells were collected for analysis of the changes of morphology,EMT related markers and tumor characteristics.Results Following indirect co-culture with hAD-MSCs,breast cancer cells morphology and EMT specific markers altered response to hAD-MSCs.The invasive and migration capacity of MCF-7 cells after co-cultured with hAD-MSCs was significantly enhanced,while no change was observed in cell cycle and proliferation.Conclusions hAD-MSCs can stimulate epithelial to mesenchymal transition of breast cancer cells.
