摘要
以烟叶蛋白为原料,用碱性蛋白酶进行水解。研究加酶量、pH值、温度、水解时间对烟叶蛋白水解度的影响。在单因素试验分析的基础上,进行正交试验,结合工业生产的实际情况,确定碱性蛋白酶水解烟叶蛋白的最佳工艺条件为:酶加量30 000 U,pH值9.0,温度55℃,水解时间2 h,在此条件下,水解度可达到24.719%。
Tobacco leaf protein is extracted from fresh tobacco leaf and hydrolyzed by Alcalase. The influences of the amount of enzyme, pH, temperature and time on the degree of hydrolysis of tobacco leaf protein are investigated. On the basis of single factor tests and the orthogonal test, the optimal working parameters are worked out and tested. To consider the feasibility in industry, the optimum hydrolysis parameters are as follows: the amount of enzyme 30 000 U, hydrolyzing 2 h, temperature 55℃, pH 9.0, and the degree of hydrolysis is 24.719% under the optimum conditions.
