川芎嗪联合丹参酮ⅡA对缺血再灌注大鼠肺损伤的保护作用研究 被引量:4

Protective Effect of Ligustrazine Combined with Tanshinone ⅡA on Lung Injury in Rats with Ischemia-reperfusion Injury

摘要 目的:研究川芎嗪联合丹参酮ⅡA对缺血再灌注大鼠肺损伤的保护作用。方法:实验分为空白对照(等容生理盐水)、假手术(等容生理盐水)、模型(等容生理盐水)、地塞米松(5mg.kg-1)和川芎嗪+丹参酮ⅡA高、中、低剂量(40+12、20+6、10+3mg.kg-1)组。于肺缺血前3d灌胃给药,每天1次。测定大鼠血浆丙二醛(MDA)含量、肺匀浆髓过氧化物酶(MPO)活性、湿重/干重比值(W/D)和肺通透指数。结果:与空白对照组比较,模型组大鼠血浆MDA含量显著增加(P<0.01),肺匀浆MPO活性显著增强(P<0.01),W/D、肺通透指数显著增加(P<0.01)。与模型组比较,川芎嗪+丹参酮ⅡA高、中剂量组大鼠血浆MDA含量显著减少(P<0.01或P<0.05),肺匀浆MPO活性显著减弱(P<0.01或P<0.05),W/D、肺通透指数显著减小(P<0.01或P<0.05)。结论:川芎嗪+丹参酮ⅡA联合用药对肺缺血再灌注大鼠有一定的保护作用,其机制与降低过氧化物代谢产物、减少中性粒细胞生成、增加肺和毛细血管的通透性有关。 OBJECTIVE: To study the protective effect of ligustrazine combined with tanshinone Ⅱ A on lung injury in rats with ischemia-reperfusion injury. METHODS: SD male rats were divided into 7 groups, i.e. blank control group (constant volume of normal saline), sham operation group (constant volume of normal saline), model group (constant volume of normal saline), dexamethasone group (5 mg.kg-1), TMP+tanshinone Ⅱ A high-dose, medium-dose and low-dose groups (40±12 mg.kg 1, 20+6 mg- kg-1, 10+3 mg.kg-1). Those groups were given relevant medicine intragastrically once a day 3 days before ischemia. MDA content, MPO activity, wet weight/dry weight ratio (W/D) and pulmonary permeability index were determined. RESULTS: Compared with blank control group, MDA content of model group increased significantly (P〈0.01), MPO activity of lung homogenate was significantly enhanced (P〈0.01), W/D, pulmonary ventilation index increased significantly (P〈0.01). Compared with model group, plasma content of MDA in TMP+tanshinone Ⅱ A high-dose and middle dose groups was decreased significantly (P〈0.01 or P〈0.05), lung homogenate MPO activity was significantly decreased (P〈0.01 or P〈0.05), LF/D and pulmonary ventilation index decreased significantly(P〈0.01 or P〈0.05). CONCLUSION: Combined therapy of lignstrazine+tanshinone Ⅱ A has a protective effeet on lung ischemia-reperfusion rats, its mechanism is related to reduction of peroxides metabolites, reduction of neutrophils generation, increase of pulmonary capillary permeability.
