摘要
目的研究Nogo-A基因3'-非翻译区单核苷酸多态性(SNP)各等位基因及基因型在中国广西地区人群中的分布频率,比较其在不同种族间分布的差异。方法采用单碱基延伸的PCR技术和DNA测序法检测199例中国广西人的Nogo-A基因3'-非翻译区rs2588510C/T、rs887G/A多态性,并结合人类基因组计划(Hapmap)公布的四个人群(欧洲人、非洲人、日本人和中国北京人)的SNP分型数据,比较分析这五个人群的基因型及等位基因的分布频率。结果在中国广西人群中存在Nogo-A基因3'-非翻译区rs2588510C/T、rs887G/A多态性。与欧洲及非洲人群相比,Nogo-A基因多态性的分布频率均存在显著性差异(P<0.05);而与日本人及北京人相比,差异无显著性(P>0.05)。结论在中国广西人群中存在Nogo-A基因多态性,但与其他种族人群比较存在显著性差异,这种差异可能是导致一些疾病在不同种族间的发病率和临床表现存在显著不同的因素之一。
Objective To study the frequencies of allele and genotype distribution of Neurite growth inhibitor-A(Nogo-A) gene Y-untranslated regions single nucleotide polymorphism(SNP) in Chinese guangxi populations, and to analyze the distribution of Nogo-A polymorphism among different races. Methods The Nogo-A gene 3'-untranslated regions rs2588510 C/T and rs887 G/A polymorphisms were examined by the polymerase chain reaction-single base extension(PCR-SBE) technique and DNA sequencing methods in 199 Chinese Guangxi people. Frequencies of alleles and genotypes of Nogo-A gene rs2588510 C/T, rs887 G/ A polymorphism in Guangxi population were analyzed, compared with the other four population(HapMap- CEU, HapMap-YRI, HapMap-JPT and HapMap-HCB) from Human Genome Project group(Hapmap) data. Results There was Nogo-A gene polymorphism in Guangxi population. The frequencies of allele and genotype distribution of Nogo-A gene rs2588510 C/T, rs887 G/A polymorphism were significantly different, compared with HapMap-CEU and HapMap-YRI populations(P〈0.05), the Nogo-A gene polymorphism were not significantly different, compared with HapMap-JPT and HapMap-HCB(P〉0.05). Conclusion There were Nogo-A gene polymorphism in Guangxi population, the frequencies of polymorphism distribution of Nogo-A gene were significantly different, compared with other ethnic populations. The variation of Nogo-A gene polymorphism among different ethnic groups might account for the varied clinical spectrum of some Nogo-A related diseases.
