摘要 用紫外线照射枯草芽孢杆菌 (Bacillussubtilis)C1,获得了一个莽草酸营养缺陷型变异株。与亲株相比 ,该菌株完全丧失了转酮醇酶活性 ,能够在培养基中大量积累D 核糖。在30L的发酵罐上培养 6 8h ,D 核糖的产量为 6 0 9g/L。发酵液经离子交换树脂纯化后 ,获得了发酵产物结晶。发酵产物的红外光谱分析与标准样品一致 ;纸层析显色后呈单一斑点 ,其Rf值与标准样品相同。 Bacillus subtilis C1 was mutagenized by using ultraviolet irradiation. A mutant which required shikimic acid for it's growth was isolated.The mutant had entirely lost transketolase activity and could accumulate a large amount of D? ribose in the culture medium. The amount of D? ribose was 60.9?g/L of the broth incubated in 30?L fermentor for 68?h. The product was purified from the fermentation broth with ion exchange chromatography and identified as D? ribose by means of infrared absorption spectrum and paper chromatography.
