摘要 目的探索人胚干细胞来源的成纤维细胞(EBF)作为纳米银颗粒细胞毒性评价模型的可行性,旨在为纳米材料生物安全性评价提供新的细胞模型。方法将纳米银与EBF细胞共孵育,采用相差显微镜、CCK-8法、流式细胞术和透射电子显微镜分别对细胞形态、细胞增殖、细胞周期及细胞摄取进行研究。结果纳米银颗粒对EBF活性的影响是浓度和时间依赖性的,纳米银可以进入细胞内并引起细胞周期阻滞。结论 EBF可以作为一种新的纳米银颗粒生物安全性评价模型,由于其来源的特殊性,可进一步模拟人体正常细胞的反应。 Objective To explore the feasibility of human embryonic stem cell-derived fibroblasts(EBF) to be as a novel cell model to evaluate the cytotoxicity of silver nanoparticles(Ag-NPs).Methods After exposing to Ag-NPs the cell morphology,cell proliferation,cell cycle and cellular uptake were examined by phase-contrast microscope,CCK-8,flow cytometry and transmission electron microscope.Results The cytotoxicity of Ag-NPs in EBF was dose and time-depedent.Ag-NPs can enter the cells and induced a G2/M arrest.Conclusion EBF could be taken as a valid candidate for cytotoxicity screening assays of nanoparticles as this model achieves a closer representation of the in vivo response.
