摘要
目的:用HPLC法测定清热化毒丸中黄芩苷的含量。方法:采用高效液相色谱法,使用Agilent ZORBAX SB-C18(250mm×4.6mm,5μm)色谱柱,以甲醇-水-磷酸(47∶53∶0.2)为流动相,检测波长为315nm,流速:1.0mL·min-1,柱温:40℃。结果:黄芩苷在0.1028~1.0275μg范围内呈良好的线性关系,回归方程为y=10225920x-1083344(r2=0.9925)。平均回收率为99.61%,RSD=1.4%。结论:本法灵敏、准确、专属性强,回收率高,重现性好,可作为清热化毒丸中黄芩苷的含量测定的检测方法。
Objective: To establish the method for determining berberine hydrochloride in Qingre Huadu Wan by HPLC. Methods: Berberine hydrochloride was analysed simultaneously by HPLC. The optimal conditions of separation and detection were achieved on a Agilent ZORBAX SB - C18 (250 mm ×416 mm, 5μm ) by isocratic elution with the mobile phase of acetonitrile - water (47: 53 ) at the flow rate of 1.0 mL · min-1. The detective wavelength was 315nm. Results: The calibration curves of berberine hydrochloride showed that the linearity were completely acceptable. The average recovery of berberine was 99. 61% respectively. The RSD wasl. 4%. Conclusion: This method was simple, rapid and feasible for the quantification of berberine hydrochloride in Qingre Huadu Wall.
