摘要
利用双向凝胶电泳和基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)技术,研究与人肺永生化支气管上皮细胞(BEP2D细胞)辐照相关的差异蛋白质,寻找辐射致肺癌早期诊断和治疗的新指标。结果表明,提取样品蛋白进行二维凝胶电泳,找到差异表达蛋白质点约18个,质谱鉴定获得10张肽质指纹图,经数据库搜索后初步鉴定差异表达的蛋白质有7种,即亲环素A、肌球蛋白轻链2、细胞角蛋白9、α-烯醇酶、磷酸丙糖异构酶1、MKRN1、c-myc启动子结合蛋白1。该7种蛋白质与辐射诱导肺癌的发生、发展过程密切相关。
The samples of the BEP2D cell radiated with 0、1.5 and 3.0 Gy γ rays were cultivated and collected,and the total protein were separated with the two-dimensional gel electrophoresis.The peptide mass fingerprint were obtained using matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS)for differentially expressed proteins.10 peptide mass fingerprinting are reveal in the mass spectra identification,8 of them are approved in the database,and 1 are duplicated;2 are null. There are 7 points identified in the initial testsby Mascot software in the NCBInr database,such as Cyclophilin A,Myosin light chain 2,Cytokeratin 9,α-enolase,Triosephosphate isomerase 1,Makorin ring finger protein 1,c-myc promoter-binding protein 1.these 7 kinds of proteins probablely are the special proteins related to the lung cancer induced by irradiation,which might be worth of further study of the functions of proteins.
