摘要 目的观察胰岛素样生长因子-1受体(IGF-1R)和基质金属蛋白酶-2(MMP-2)两种蛋白在肾细胞癌组织中的表达并探讨其临床病理意义。方法应用免疫组化两步法检测72例肾癌组织和33例同期癌旁正常肾组织中IGF-1R和MMP-2蛋白的表达水平,分析IGF-1R和MMP-2与肾细胞癌病理类型、分级、分期及肿瘤发生、发展和转移之间的关系。结果肾细胞癌组织中IGF-1R和MMP-2的表达率分别为69.4%(50/72)和72.2%(52/72),明显高于同期非肿瘤癌旁正常肾组织中的39.3%(13/33,P<0.05)和27.3%(9/33,P<0.05),两种蛋白在肾细胞癌组织中的表达水平均与患者的肿瘤大小、淋巴结转移、病理分级和临床分期存在相关性(P均<0.05),与肿瘤的病理分型、患者年龄、性别无相关性(P>0.05);IGF-1R和MMP-2两种蛋白的表达呈正相关(P<0.05)。结论 IGF-1R和MMP-2与肾细胞癌的发生、发展和转移过程密切相关,同时检测这两种蛋白的表达可以对肾细胞癌恶性程度的判定及进一步治疗提供参考依据。 Objective To observe the expression of insulin-like growth factor-1 receptor(IGF-1R) and matrix metalloproteinase-2(MMP-2) protein in renal cell carcinoma and to investigate their clinicopathological significance.Methods Expression of IGF-1R and MMP-2 protein in 72 specimens of renal cell carcinoma and 33 non-tumor renal tissues were examined by using polymer detection system for immuno-histological staining and crosscompared with clinicopathological features of renal cell carcinoma.Results High expression of IGF-1R and MMP-2 protein were observed in 69.4%(50/72) and 72.2%(52/72) of renal cell carcinoma tissues respectively,significantly higher than those in non-tumor renal tussues 39.3%(13/33,P0.05) and 27.3%(9/33,P0.05).The high expression rate of IGF-1R and MMP-2 were significantly associated with the tumor size,lymph node metastasis,Fuhrman classification and Robson staging(allP 0.05),which had no relation to the tumor pathologic types,patient age,gender(all P 0.05).The expression of IGF-1R was positively correlated with the expression of MMP-2 in renal cell carcinoma(P0.05).Conclusion High expression of IGF-1R and MMP-2 protein are significantly correlated with the creation.The development and the metastasis of renal cell carcinoma,examination of expression of IGF-1R and MMP-2 may be useful for judgement of maligancy of renal cell carcinoma and reference of further therapy.
